An Efficient Agrobacterium-Mediated Genetic Transformation Method for a Monocot Model Plant, Brachypodium distachyon

Model plants are necessary and important for advances in various research areas of plant biology. Compared with Arabidopsis thaliana, a dicot model that is widely used thus far, monocot not popular using biology research. In this regard, Brachypodium distachyon suggested as new alternative economically valuable temperate grasses cereals such wheat barley, also bioenergy crops switchgrass miscanthus. One the major constraints performing would be difficulty genetic transformation its low frequency. Here, we provide detailed improved method Agrobacterium-mediated B. distachyon, based on our experimental experience. With hygromycin resistance selectable marker, could obtain approximately 40%−60% efficiency method, which process takes 26 weeks to T1 seeds starting from co-cultivation immature embryos Agrobacterium cells. describe about factors possibly contributing distachyon.
